Dear All,
I have a non-profit customer that would like to have polyclonal antibodies made
in New Zealand white rabbits and purified. The antigen has been used
successfully in the past, and the polyclonal antibodies produced have been
marketed. Recently this customer has attempted to generate polyclonal
antibodies with a commercial entity. The attempt was unsuccessful. The customer
believes that they will have better results working with a university core
facility.
They would like to use the same animals and immunization routine that yielded
enantiomer specific antibodies. The polyclonal antibodies work with WB of cell
lysates and bind free hapten. The antigen is a hapten conjugated to KLH using
glutaraldehyde. The customer will provide the antigen.
Here is the immunization scheme:
* Freund's Complete Adjuvant was used for the primary immunization, and
boosts consisted of antigen alone.
* 0.5 mL FCA + 500ug antigen in 0.5 mL
* 0.25 mL injected into 4 sites SQ
My core does not work with rabbits. I am looking for an experienced group that
can move forward with this project quickly and be prepared to boost and collect
sera monthly from animals for 10 months using the protocol above. The screening
of the sera would be done by the customer prior to purification. The
purification would be done in your facility. In the past, this group used
ammonium sulphate precipitation methods. They are amenable to trying Protein
A/G columns.
